The Mexican international has been out for four weeks after injuring his shoulder in an awkward fall against West Ham.
But the 23-year-old attacking midfielder returned to full speed work with the first team yesterday and is raring to get back into action.
After paying part of his own 12-month loan fee to come to City from Ukraine side Shakhtar Donetsk, Castillo made only three starts before being left sidelined and frustrated.
"The medical people say Nery is now allowed to do everything with the senior squad including physical contact and tackling, so that is good," reported manager Sven-Goran Eriksson.
"It has not been easy for him having to work with the fitness coaches all the time."
Meanwhile, China international Jihai Sun has praised Eriksson for his inspirational team talk before Sunday's derby win.
Sun said: "It was a brilliant feeling. Before the game, the manager told us that it was 34 years since City had won at Old Trafford and that we could go out and make history.
"He said that we could be heroes and people would remember us in 34 years time if we won.
"Seeing the fans when I went out to warm-up brought his words back to me, and I thought to myself `Jihai, you can be a hero here'. To win like we did was a fantastic feeling.
"The atmosphere in the dressing room after the win was amazing, everyone was singing. It's one of the best moments of my City career without a doubt."

I think when we have all our players fit (soon) then when we play the lower teams & home games you might see 442!
But not bothered where nery plays as long as he gets on the pitch! Although Vass scored on sunday I think I would prefer Nery on the righ cutting in & Pets on the left banging in crosses.

At the moment, on the bench. But we pratically have a full fit squad at the moment, which is almost unheard of.
We will get injuries in the second half of the season.
He can fill in for Petrov for example, if he gets injured or loses form. Ditto Benjani.
Our squad and bench is looking very strong at the moment.

He looked a bit lightweight, but he'll need time to adapt & hopefully his speed will be an asset to the team whereever Sven decides to play him.

Anyone who saw Castillo in the copa America will know he's quality. But I think he's one of those that will take a while to adapt to the premiership. So don't expect to see the best of him till next year. His best position for us at the moment will probably be in the Vass role or alternatively as an attacking sub playing just of the front man. It'll all create a healthy competition for places, if nothing else.
