WBA light-welterweight champion, Khan, has set his sights on Erik Morales or Juan Manuel Marquez after defeating Andreas Kotelnik last month.
But Witter was hoping for a unification fight if he claimed the vacant WBC title against Devon Alexander on Saturday - insisting he would knock Khan out inside three rounds.
But his failure to come out for the ninth round against Alexander - because of an arm injury - has just about killed his hopes of facing Khan.
It is another disappointment for Witter, who for so long dreamed of a mega-money payday against Ricky Hatton.
But his hopes of meeting up with Khan were always far-fetched with the Bolton fighter now hoping to make his mark in America by fighting some of the biggest names State-side.
The 22-year-old is taking his time before deciding on his next opponent, but had hoped on a different battle of Britain against Hatton.
The Hitman is still deliberating over whether to return to the ring after defeat to Manny Pacquiao in May and has distanced himself from a clash with Khan.
That has left Frank Warren and Freddie Roach weighing up the next step for Khan, with Morales featuring high on their short-list.
And now that he is a world champion, Khan says he is willing to be patient in quest to become the best pound for pound fighter in the world.
Improve
"In the early days I used to rush," he said. "The first thing Freddie said was 'think about what you do. Don't get caught.'
"The knockdowns I had in my early career were because I went onto the shots, which made them harder.
"I can improve and get better and better. This is a big step - I'm now at the world class stage and want to get better.
"I will have a few weeks off and set my goals and ambitions.
"It's time for me to go to America and fight with the best.
"Frank Warren is a great promoter. Next thing I want to do is fight the best out there."
Without the WBC title, Witter does not fit that description, but he insisted he had no option but to retire against Alexander.
"My elbow went in round four and basically I couldn't keep him off with my left hand," said Witter.
"I battled through for a few rounds but the pressure kept coming on and it was like 'I can't keep doing it.'"
Manchester featherweight Terry Flannigan improved to 3-0 as a pro at Colne's Town Hall by registering a 40-35 four round points win over Middlesbrough's Michael O'Gara.
Manchester light-welter William Warburton earned a share of the spoils in his six-rounder with unbeaten Burnley puncher Chris O'Brien, Fleetwood referee Steve Gray scoring the bout 57-57.

Are you serious? He's nothing untill he fights him again. He's calling out people, why's he not calling out Prescott if he's learned from his mistakes? Freddie Roach and Frank Warren will be hand picking opponents from now on as usual they made a mistake with Prescott and will not be going back.

My point is he's bigging himself up. I could say the same to you,'so what' Marquez beat Prescott, doesn't mean Khan is better than Prescott if Khan then beats Marquez. Do you work for a certain promotion company "A rematch may happen in the future but not yet" you mean when its known Prescott is having money troubles and fitness issues you(the compnay) will pay him to fight Khan at short notice. I could go on,,but I give up. Khan ran from a non puncher in his last fight. None of us like getting hit, but he is really scared of getting hit as he knows he can't take it

I didn't even watch the fight at the time but as soon as I heard I said to the people I was out with that I would bet he'd been banged with a left hook to the chin and hey presto that was the shot that did the damage....and then the Snowman came on.
I like Amir, but I think Politics and being more Marketable have excelled the lad and ground out the financial success that his ability alone might not yet justify. I understand that it's a business and promoters and media have their own interest at the centre of everything they do, but JM would beat him but look at him, not had the same exposure or financial success etc
Good luck to both of them, local lads etc: but I fancy JM if this fight ever happens.

The Khan that couldn't take one on the chin is now very scared of taking one on the chin.I've watched Khan for a long time and the only difference from the school boy champ and the young man that beat Kotelnik was his left hook to the body, he's mastered that quite well but it didn't hurt Kotelnik!!??and when Kotelnik landed a packet of crisp on Khan in round 10 he ran like a rabbit from a greyhound
