The one-hour service to London City Airport is due to take off on September 9 and will be operated by Netherlands-based VLM Airlines, using a Fokker 50 aircraft.
Flights will leave Manchester at 06.40, 09.30, 12.30, 15.30 and 18.30 and turnaround times in London are scheduled to be half an hour.
One-way fares will start from £20 (plus taxes) and VLM regional manager Titus Johnson said: "The service will offer business passengers the fastest route in and out of London."
