The Rt Rev Mark Davies, Bishop of Middleton, was one of eight clergy who prayed behind the glass as passers-by looked on.
It was part of a continuous prayer session run by Sanctus1, an alternative church based alongside the Nexus Art Café in Dale Street, Manchester, to mark the 40 days of Lent and highlight loneliness.
Members of the public are also being invited to spend time in the window.
The Rev Ben Edson, leader of Sanctus1, said: "The city can be a place where we feel like we're in a glass box where all the action is happening around us and we're just an invisible bystander.
"The 40 Days exhibition aims to highlight the loneliness and anonymity of the busy city."
