The Commonwealth lightweight champion will be the main attraction on a show to be staged at the 21,000-capacity Arena on September 6, a bill which promoter Frank Warren has promised will be a "blockbuster."
The fight is likely to be Khan's temporary farewell to Britain, as he is expected to head to the USA later in the year for experience and exposure to an American audience.
An opponent has yet to be announced, but high on the list of possibilities will be the winner of the July 11 British lightweight title showdown between Manchester's John Murray and Salford-born Lee Meager.
Khan has been urged to stay at domestic level for the time being after he was downed and almost shocked by another Manchester fighter, Michael Gomez, at the weekend.
And the fact that the show has now been confirmed for the MEN Arena heightens the chances of Murray or Meager getting a shot, as a `derby' contest would excite local interest, as well as making the fight a unification contest for the Brtish and Commonwealth belts.
Arthur is the WBO super-featherweight champion, and will be making the second defence of his title against an opponent to be announced.
The show could also feature the debut of giant heavyweight prospect Tyson Fury, from Styal, who has exclusively announced to M.E.N. Sport that he is about to turn professional.
